DX243 shows sustained neuroprotection of key auditory circuits involved in Age-Related Hearing Loss.

Dendrogenix reports the publication of new preclinical results highlighting the ability of DX243 to protect the most vulnerable auditory circuits from age-related degeneration.

Published in Frontiers in Pharmacology (2026), this study from the Tübingen Hearing Research Centre (University of Tübingen, Germany) demonstrates that a 14-day daily treatment with DX243 preserves the functional integrity of critical auditory synapses involved in temporal sound processing and speech understanding in noisy environments—capabilities that decline early in Age-Related Hearing Loss (presbycusis).

Protective effects persist for up to 6 weeks after treatment cessation, supporting a durable disease-modifying effect on cochlear synaptopathy rather than short-term symptomatic relief.

Unlike conventional strategies aimed at global neuronal survival, DX243 selectively targets cochlear synaptopathy, a mechanism increasingly recognized as a central driver of age-related auditory dysfunction. Dose-dependent preservation of synaptic integrity and neurophysiological responses further confirms a strong and consistent neuroprotective profile at both peripheral and central levels.

By demonstrating that early pharmacological intervention can preserve the auditory fibers most sensitive to cumulative acoustic insults, this publication positions DX243 as a promising, first-in-class candidate for the prevention and/or treatment of age-related hearing loss, addressing a significant unmet medical need and paves the way for a new class of therapeutic treatment to slow or modify the natural aging of the auditory system.
